is there a way to create a door that needs an item to open it?
Try to use a vending machine (requires a specific item in order to purchase), wire it to a barrier that deactivate when item purchased.

For memory efficiency, you could use a button and a checker. Of course what Bird and NotYoyo said are very good methods, but if you want to save memory. Remember if you find a solution be sure to mark it, so that the topic won’t become to much of a clutter.
